Petty thief Nathan Drake is recruited by fortune hunter Victor Sullivan in an adventure to locate the lost treasure of the Magellan expedition, a quest that caused Nathan's long-lost brother Sam to mysteriously disappear. However, corrupt billionaire Santiago Moncada (Antonio Banderas) is also determined to reach the treasure first and has recruited unscrupulous mercenary leader Jo Braddock (Titi Gabrielle) to locate Magellan's hoard at any cost.
